Better payload decisions with Bio‑Courier® TL

Conventional transfection reagents often compromise cell viability, introducing biological noise. Bio‑Courier® TL keeps cells viable, so what the assay measures is the payload. And because the same chemistry carries that payload into animal studies, it can be evaluated in vivo without the confounding effects introduced by a different carrier chemistry.

Cell toxicity creates payload-independent artefacts that confound the readout.

In animals as well as in cell culture

  • Extrahepatic delivery

    Delivery beyond the liver

  • Multiple administration routes

    Flexibility in study design

  • Ease of use

With the carrier held constant, in vitro data are a better indicator of in vivo performance.

Why Bio-Courier® TL works both in vitro and in vivo.

Bio-Courier® TL stabilises lipids with a silicon matrix and requires less cationic charge than other reagents. This results in effective transfection with less toxicity in both cell culture and in animals.

Cationic transfection reagents: the culture dish

One high cationic charge does two jobs: it condenses and holds the nucleic acid, and it drives cell entry. Charge scales with payload, and the charge that delivers is the charge that disrupts membranes. Efficacy and toxicity are coupled, which is what keeps these reagents in the dish.

Ionisable lipid nanoparticles: the systemic route

Ionisable lipids bind the payload tightly and release it only once the endosome acidifies. That same tight binding limits release inside cultured cells. The chemistry is tuned for the bloodstream, not the culture plate.

Bio-Courier®: both

A silicon matrix does the structural work: it stabilises the particle and holds the nucleic acid. The cationic lipid is then left with one job — providing the surface that meets the cell — so far less cationic charge is needed for efficient transfection. Less charge means the toxicity that comes with charge does not follow the reagent into the animal.

Bio-Courier® TL: the in vivo advantage

In vivo, Bio-Courier® TL offers numerous advantages over other in vivo reagents and conventional LNPs.

  • Unlike conventional LNPs, which are formed around their cargo, Bio-Courier® TL is a finished empty nanoparticle: the payload is loaded afterwards, in under an hour at room temperature, at above 95% encapsulation efficiency.
  • The formulations need no ultra-cold chain.
  • Multiple administration routes: intravenous, subcutaneous, intraperitoneal, intramuscular, oral, dermal and eye-drop delivery have all been demonstrated.
  • Reduced hepatic accumulation, and delivery to other organs and tissues.
  • Bio-Courier® TL is well tolerated: more than 1,300 animals have been dosed without adverse reactions.
  • In addition to non-GLP in vivo studies, Bio-Courier® TL provides a defined path to IND-enabling studies and the clinic.
